PCR is a technique for rapidly replicating DNA in vitro.The DNA double strand will be untied at high temperature, and the primer will bind to the DNA single strand at the appropriate annealing temperature.When the temperature rises again, DNA polymerase will use DnTP to synthesize the complementary DNA strand rapidly.After dozens of cyclic reactions, the target DNA product can grow exponentially.

After the PCR reaction system is prepared, a special PCR centrifuge can be used for centrifugation before placing it in the PCR instrument, which can avoid wall hanging and remove some bubbles.

I. Reagent Preparation Area

A mini centrifuge (suitable for 1.5 ml, 0.5 ml, 0.2 ml centrifuge tubes and 0.2 ml for PCR with 8 rows of centrifuge tubes) is need to centrifuge reagent.

II.Sample Preparation Area

1)A table high speed centrifuge is needed for nucleic acid extraction of DNA samples.

2) A table high speed refrigerated centrifuge is needed for RNA sample nucleic acid extraction.

3) A table low speed centrfuge is needed for blood serum separation.

Because the PCR process is to detect the specific fluorescence intensity of each site. If sufficient mixing and centrifugation is not carried out before PCR, first, the amount of reaction reagents may be different, the uniformity is not good, affecting the fluorescence intensity.Second, if there is a bubble, it will refract and reflect the fluorescence, and it will also block the light path.

To sum up, it is necessary to mix thoroughly before PCR and centrifuge without bubbles.
